Research: In sedentary mice, four weeks of AICAR alone raised treadmill running endurance by 44% (Narkar et al, Cell 2008;134(3):405-415, PMID 18674809). The only large human trial tested acadesine, its nucleoside form, in 3,080 patients having bypass surgery and was stopped for futility: the composite of death, stroke or severe left-ventricular dysfunction occurred in 5.1% on acadesine and 5.0% on placebo (RED-CABG, Newman et al, JAMA 2012;308(2):157-164, PMID 22782417). The mouse endurance result has never been tested in people.
Side effects: No adverse event table for AICAR as sold has been published, RED-CABG reported no difference from placebo in its main efficacy and safety end points for infused acadesine
